The word ‘rational’ is properly used to talk about cognitive algorithms which systematically promote map-territory correspondences or goal achievement.
I disagree with the definition “systematically promote map-territory correspondences” because for me it is “maps all the way down”, we never ever perceive the territory directly, we perceive and manipulate the world via models (maps). Finding models that work (that enable goal achievement/winning) is the essence of intelligence. “All models are wrong, some are useful”. Even if we get to the actually elemental parts of reality and can essentially equate our most granular map with the territory that is out there, we still mainly won’t care in practice about this perfect map because it is going to be computationally intractable. Lets take Newtonian Mechanics and General Relativity for example. We know that General Relativity is “truer” but we don’t use it for calculating pendulum dynamics at the earth surface, the differences that it models are just irrelevant compared to other more relevant stuff.
Second: I’m mostly making an empirical claim as to what seems to happen to individual people (and more noticeably to groups-of-people) if they focus on the slogan “rationality is winning.”

That’s why I give examples of areas where the feedback loops are short, fast and with very little noise. These are examples that worked for me with astonishing efficiency. I would be the person I am otherwise. And I’ve chosen these areas explicitly for this reason.
If you set out to systematically win, many people end up pursuing a lot of strategies that are pretty random. And maybe they’re good strategies! But bucketing all of them under “rationality” starts to deflate the meaning of the word.

Here I make an extremely strong claim that it is never rational to believe false things. Personal integrity is the cornerstone of rationality and winning. This is a blogpost scope topic, so I won’t go into it further right here.
Similarly and more specifically: a lot of things-that-win in some respects are wooy, and while I think there’s in fact good stuff in some woo, while the first generation of rationalists exploring that woo were rationalists with a solid epistemic foundation. Subsequent generations came more for the woo than for the rationality (See Salvage Epistemology).
“Woo” is stuff that doesn’t fit into your clear self-consistent world model. There is a lot of useful stuff out there that you guys ignore! Copenhagen interpretation, humanities, biology, religion, etc… If you don’t understand why it makes sense, you don’t understand it, fullstop. I believe that mining woo for useful stuff is exactly how you do original research. It worked wonders for me! But integrity goes first! You shouldn’t just replace your model with the foreign one or do “model averaging”, you should grok what those guys get that you are missing and incorporate it in your model. Integrity and good epistemiology are a must, if you don’t have those yet, don’t touch woo! This is power aka dark arts, it will corrupt you.

Third: The valley of bad rationality means that study of systemized winning is not guaranteed to actually lead to winning, even en-net over the course of your entire lifetime.
In my experience there is a valley of bad X for every theory X. This is what you have to overcome. I agree that many perish in it. But the success of those who pass is well worth it. I think we should add more “here be dragons” and “most of you will perish” and “like seriously, 90% will do worse of by trying this”. It’s not for everybody, you need to have character.
Fourth: Honestly, while I think LessWrong culture is good at epistemics, addressing motivated cognition, and some similar things… I don’t have a strong reason to believe that we are particularly good at systematically winning across domains (except in domains where epistemics are particularly relevant)

A lot of my answer here is “sure, that might be fine!” I highly recommend you focus on winning, and use whatever tools are appropriate, which sometimes will be “study/practice cognitive algorithms shaped” and sometimes will have other shapes.
I do agree there is a meta-level skill of figuring out what tools to use, and I do think that meta-level skill is still pretty central to what I call rationality (which includes “applying cognitive algorithms to make good decisions”). But it’s not necessarily the case that studying that skill will pay off.
Linguistically, I think it’s correct to say “the rational move is the one that resulted in you winning (given your starting resources, including knowledge)”, but, “that was the rational move” doesn’t necessarily equal “‘rationality’ as a practice was helpful.”

I see from the 5th point that you explicitly included it, sorry for missing it, I just tend to really get stuck in writing good deliberate replies, so I just explicitly decided to contribute whatever I realistically can.
I still stand on the position that this one (I call it critical thinking) should come first. It’s true that there is no guarantee that it would pay off in time for everybody. But if you miss it, how do you distinguish between woo and rationality? I think you are just doomed in this case. Here be dragons, most of you will perish on the way.
True (Scottish) Rationality is winning. Firstly, whom do we call a rational agent in Economics? It is a utility optimiser given constraints. An agent that makes optimal choices and attains max utility possible, that’s basically winning. But real life is more complicated, all agents are computationally constrained and “best case” is not only practically unattainable, it is uncomputable, so we cannot even compare against it. So we talk about doing “better than normally expected”, “better than others”, etc. When I say “winning” I mean achieving ambitious goals.
But achieving ambitious goals in real life is mainly not about calculating the optimal choices! It is mainly about character, integrity, execution, leadership and a lot of other stuff! How come I still claim that Rationality is Winning? What use is knowing what to do if in practice you don’t do it? Well, that’s the point! An “optimal” strategy that is infeasible is not optimal)
But why focus on rationality at all if other stuff is more important? Because, well, your character, integrity, execution, resources, etc are not under your direct control except via the decisions that you make. You get them by making rational decisions. Making decisions that make you win and achieve your (ambitious) goals is what I call rationality.

Why do you exclude one of the most important cognitive algorithms “sifting out the good from the bad” from “the study (and applied skill) of finding cognitive algorithms that form better beliefs and make better decisions”? If you are not good at critical thinking, how do you know that LW is not complete bullshit?
“Developing rationality” as a goal is an awful one because you don’t get feedback. Learning doesn’t happen without feedback. “Winning” may be a great one IIF you pick fields with strong immediate practical unbiased feedback. For example, try playing poker, data science in an executive position, all kinds of trading (RTB, crypto, systematic, HFT, just taking advantage of the opportunities when they present themselves), doing research just for the purpose of figuring out stuff for yourself because your life & money (universal resource) depends on it (or because it’s fun and later your life will depends on it :) ). These are all examples from my life and they worked wonders for me.

How did you get the implied probabilities from the vanilla option markets? I don’t know an obvious way to do it, perhaps the simplest (and wrong) approximation would be to take the BS IV of traded vanillas at the strike price and plug it into binary BS formula?

Theoretically, it shouldn’t cause an immune response, as peptides shouldn’t be immunogenic on their own, that’s why you need chitosan as a delivery mechanism and adjuvant. However, who knows? Was it actually researched and proven that peptides on their own do not cause an immune response no matter how big is the dose and route of administration? I could well imagine that this is simply a theoretical conclusion that was never empirically verified, or that it was only verified by an injection, but not by snorting, and peripheral immune system is triggered by pure peptides, while systemic is not, or that it was only tried in much lower doses. Even if a 10-100 times higher dosage of peptides is equivalent to chitosan+peptides, this is likely of little commercial interest as chitosan “enhancement” is cheaper and more scalable at commercial scale than peptides production. So it might actually work and it is cool that you’ve tried it and there is evidence that “you were congested for a few days like after previous vaccine applications and it looks like nothing really bad had happened”.
What dose of pure peptides did you use per 1 peptide and totally? The currently recommended dose is ~200ug in total, no matter how many component peptides are there. If you do 5 peptides like in the v10, then this is 40ug per peptide. If you do 1mg=1000ug of pure peptide, then this is 25 time more dakka and it might well trigger the same response as 40ug of peptide+chitosan.
I think the currently recommended does is likely to be too low for average human risk preferences. Organisations are extremely risk averse, the negative side effects are given much more weight than unrealised benefits, so the recommended dose is likely to be “the smallest one that kind of works without any side effects” rather than the optimal one. There is almost no evidence for the efficiency of different doses and this is all guesswork based on guesswork of others in slightly related cases, e.g. the 500ug cancer peptide vaccine dosage.
Congestion after vaccine/peptides administration could be due to many factors, so I consider it only a very weak evidence. This can be purely psychosomatic. The vaccine is acidic, so it irritates your nasal mucosa. When you put anything in your nose, it may irritate it and cause congestion. You have applied pure peptides after receiving several doses of vaccine and after enough time has passed for developing an immunity, so congestion may indeed be an immune response, but it may only happen when you already have an immunity, but it won’t be triggered if you have no immunity. Actually, this is what your data suggests. “Previously, on doses 3-6 of the vaccine, I had consistently been congested for a couple days after”. I understand this as you having no congestion after doses 1-2 and getting it after all further doses. If this is so, then this looks like the actually important immune response of a naive immune system happens without congestion, but the immune response of a trained immune system happens with congestion.

I haven’t. Firstly, there is no proper data, just some bits of evidence. Secondly, yep, I am pretty sure that they would get in trouble if they did anything that looked like a trial, so I assume that they stay on the safe side and well, don’t do anything like a trial.

This greatly depends on what “more dakka” and “reasonable version” means. I assume that “reasonable version” implies “doesn’t cause too much harm due to immune system overstimulation”. If “more dakka” means simply a higher dosage, then I think, that this is unlikely (5%), because 1) RaDVaC team experimented on themselves quite a bit, they received a lot of dakka, but no commercial blood test detection, 2) RaDVaC team seems reasonable enough to try this approach if it looked promising. If “more dakka“ includes stronger adjuvants (chitosan is considered a weak, but safe one), then it is much more likely (20%?), because RaDVaC team didn’t investigate those (for a reason) and it sounds plausible that you can get an immune response by irritating the immune system really, really strongly.
It is always good to have additional data, but this result is totally expected, so I don’t update much based on it. When I’ve discussed the efficiency evidence with RaDVaC team, they mentioned that:
- they have tried commercial tests and didn’t manage to get even one positive result
- they have tried ELISA to measure the blood antibody titers and got some good results (part awesome, part good, part not so good)
- they have tried ELISA to measure the saliva antibodies, there are more of them than in blood samples, but there is no methodology to translate the raw data into titers for saliva
- they focus on B-cell target mechanism, not ACE2 binding and definitely not serum antibodies, so they don’t care that much about the serum antibody titers
You are right. Hedging with a long term future makes it a very targeted bet on “the real economy, especially travel halts in X months” and this is exactly what you can expect from COVID gone bad. You can see if this is priced in by comparing spring and autumn futures prices: March vs September or I would rather say May vs December. I took a look and the market is in backwardation (longer term is cheaper than short term)! The markets expect everything to be fine, it doesn’t look like COVID disaster is priced in.
I would say that it looks like a pretty good trade. Sell May, buy Dec, hold till May expiry (or till the shock is priced in) and then close both.
/tldr short spring oil futures, long volatility, but this is not a sure bet.
I don’t think that you can get significant expected profits from betting on devastating COVID scenario. Professional market participants are not all-knowing, but neither they are ignorant. They didn’t understand that COVID is important in early 2020, but they do understand it now and they do follow the news now and they should understand what the new COVID mutations mean. It is not that difficult if you know where to look.
Don’t go for the equities. Even if you assume that this information is not priced in yet, it is not that important for the equities market. Even if the economy tanks for a year and corporate profits tank for a year, everything will recover once it over and it won’t make a difference for the share prices. The share price theoretically equals the sum of the lifetime profits discounted by the current risk adjusted interest rate. The risk adjusted interest rate is very low now, so profits 20 years ahead matter almost the same as profits in 2021, so yearly profits in a specific 2021 year don’t matter much. Equities shouldn’t tank mid-term in the scenario of COVID disaster.
Central banks and governments will compensate for the shock with expansive monetary and fiscal policies. This will bring down the interest rates even further, so asset prices should actually rise as they did in 2020, especially tech stocks, but this is a risky bet, as they might go down first due to panic.
Perhaps, your best bet is to short commodities (except gold) and oil specifically. The storage is probably filled up (please check it, I just assume that it is, but I didn’t check), so the currently produced oil should be consumed now. So if there is a demand shock, the prices should drop like they did in spring. You can make use of it by shorting the oil futures with expiries when you expect the economy to be at its lowest. I guess it is in spring.
It is also a good idea to be long volatility, so you can try buying options or VIX, but options are not a good idea for a retail investor.
If the monetary and fiscal policy is aggressive and it is going to be aggressive, you can bet on interest rates going even lower and inflation going up in a few years (but not now!), so going long long-term TIPS sounds good. The issue here is that you won’t be able to get a lot leverage as a retail investor and without leverage it is not worth it. If I could, I would buy long-dated TIPS options (is it even a thing?).
I don’t trade and have never traded any of these markets, so please take it as a random internet opinion, not as a competent investment advice.
You can order online not only peptides, but also the antibodies. So if you get sick, you can theoretically inject yourself an antibody cocktail like the one that Trump received. This is obviously not a medical advice, I haven’t even researched it in depth and don’t have a plan to do so on my own.
Regarding “how do you verify that it works?”, specifically regarding the concerns whether the generated antibodies will bind to the virus and antibody enhanced disease concerns. , Right now there is a $600 test selling online (at a reputable source) that can verify whether the antibodies are neutralizing (=block the virus replication). It tests whether the generated antibodies bind to the RBD (Receptor Binding Domain) of the virus and block it from binding to the ACE2 receptor of the human cell (this is how the virus gets into the cell). It does so in a extremely simple way: it contains RBD and an ACE2 proteins plus some stuff to detect when they bind. There exist other ways to do so, but they are more expensive and not widely available. This is how advanced the science actually is, we can commercially create somewhat arbitrary parts of human cells and viral particles for a $600 price and <1 year of development.
So you can test whether the vaccine should work via antibodies IF it elicits a sufficient immunity response measured by high antibody titer. There are already widely available antibody tests to measure the antibody titer. So if you have a candidate vaccine, you order corresponding antibodies for it (also available online), test that they block the ACE2 binding, give it to humans once you are sure that it is kind of safe (peptide vaccines are) and measure the antibody titer. If it is high enough, it is very likely that the vaccine works.
You don’t need challenge trials. You still need to check for safety, but after this is done, you can simply distribute it to people, measure their antibody response and monitor whether they get sick. If they do at a substantial rate and there are no explanatory circumstances (e.g. weak immune system, very high viral load), well, something went wrong… Otherwise, all good!
I don’t know, this is simply what they state for all peptides and this is what actually took them to deliver.
If you really want to know that it works for sure, challenge trials are not that hard. Of course, you should probably check for immune response (T-cells and antibodies) and do some animal studies before going for a challenge trial. You don’t need to infect 200 people at once, you can do one-by-one from a healthy cohort with low infectious dose so that in the worst case it works as variolation. You can get volunteers from https://www.1daysooner.org/. You can do it anonymously. You can pay people in darknet with BTC/Monero for the experiments. You can only recruit people from countries where nobody cares about your small experiments (e.g. Africa or post-Soviet Union), in some countries, e.g. Russia, there is no criminal penalty for illegal human experiments, only general responsibility for causing harm (and Russian police and courts couldn’t care less about some guy who took some stuff on his own initiative following an anonymous internet advice and ended up sick or even dead).
My bad, I forgot about testing for immunity. There are 2 testable types of immunity mentioned in the paper: antibodies and T-cells. They explicitly suggest testing for antibodies before and after. However, they say that it is going to be essentially tricky with their vaccine. Firstly, the biggest effect is expected to come via cellular immunity, not via antibodies, and testing for it is not as easy as for antibodies. Secondly, the biggest effect is expected via mucosal immune system, so even the antibodies testing should be done on a nasal wash and not on a blood sample and this is a bit tricky and less reliable. Thirdly, I guess that detection of antibodies by standard antibody assays strongly depends on a specific assay and it is hard to know if the commercial test that you have access to would detect the RaDVaC-induced antibodies. I think they mention that some of the people who took the vaccine had positive antibody tests and some had negative tests.
If you are willing to spend a bit more resources on it, you can actually test for both antibodies and T-cell response. You can create a custom ELISA assay targeted specifically at response to RaDVaC antigens. I think ordering it in a lab is a bit more difficult than peptides, but still possible as a standard service. As for the T-cells, there is ELISpot or smth like that, but it is not so easy as ELISA and much more costly as it involves collection and processing of specific T-cells.
In my case, I’ve prepared the vaccine only for a few people and verification looked too costly and unreliable to be justifiable, so I decided not to do it. I hope that RaDVaC guys did it and received antibody and T-cell immune response at least in some people, but they cannot publish the positive results, because this would be tricky on the legal side. If they did it, received negative results and still recommend the vaccine, well, then they are bad guys.
The closest real life example that I know of is RaDVaC. They are professionals in this area, but they are a group of enthusiasts, not a big institution. You can read their white paper to get a basic understanding regarding what was needed to make their vaccine. The rest of the answer is for a peptide vaccine, specifically RaDVaC vaccine, this is the only class of vaccines that I know about in details.

The most crucial information is the amino acid sequences (peptides) of the virus that are 1) crucial for its functioning, 2) immunogenic, 3) would likely look the same way alone without the neighbours, 4) possible to synthesise. As far as I understood, they got 1 and 2 from public sources (preprints and journal publications), you can follow the references to check if those are actually public. 3 probably requires some special knowledge, but even then you end up with trial and error, so you can probably do without special knowledge. Predicting 4 again requires special knowledge, but there are labs who would synthesise a custom peptide for you for $300 in 4-5 weeks. If they fail to do so, you can probably work with the lab to try some special tricks (amino acid substitution, left/right isomers).

I am no even sure that you need bio/bioinformatics know-how beyond the stuff that you can read up in the wikipedia.

You can order their synthesis in a special lab for around $150-$300 per peptide, 4-5 weeks delivery time. The other ingredients you can also buy somewhere. The equipment is trivial, you can buy the whole set for $400 and cook it in your kitchen if you wish.

No. Not even wetlab stuff, more like high school chemistry lab skills.

That’s the hard part. Probably not only for a non-professional effort. The simplest answer is you don’t. You just reason that it cannot hurt that much because you know what you have mixed and just try it.
You forgot the most important question: how do you verify that it works? The answer is that you don’t. You definitely cannot do a usual clinical trial with tens of thousands people. The challenge trial doesn’t make much sense if you are only going to use it for yourself and you friend. So probably you just say “ok, it is almost pure safe and there is a chance that it works, so I better make it and take it”. If a lot of people take it, you can have observational data. If people who take are high risk (e.g. doctors) and they don’t get sick compared to their colleagues, then you have some evidence.
There’s also a cluster of secondary questions around exactly what one could do with such a vaccine, beyond using it oneself.You can publish the recipe as RadVac did. Afaik, FDA doesn’t care or cannot do much about it. You can try giving it to other people for a minimal comp. There was another case in US where a guy tried to do so with his own vaccine. FDA sued him and I think it ended up in him having to return money to the “victims”. You can give it to your friends for free, I guess nobody would care (but I don’t live in US). You can sell it in the darknet.
I would have probably paid at most ~$5k per dose, both for myself and close people in more high risk groups. This is based on a gut estimate, not on some calculations.
But I have a better answer. I made it having about 10 close people in mind. I’ve spent smth like 2-4 full-time working weeks on research research and sourcing the ingredients plus <$1.5k (this is worth much less than my time). This ended up a marginal call, considering that I finished it quite late, but it is still somewhat justified on the basis of learning a lot of things and having an interesting experience.
I estimate its safety as very safe (roughly as safe as the commercial ones) and its expected efficiency as ~30% (90% if it ends up immunogenic enough). The expected efficiency is so low, because peptide vaccines have low immunogenicity. Thinking about it now, adding an adjuvant like alum would be a good idea.
